| Genomic DNA was prepared from various blood samples using the JETQUICK Blood DNA Spin Kit. Aliquots were run on a 1% TAE agarose gel. First and last lane is the length standard Lambda HindIII/EcoRI. From left to right: EDTA-blood/fresh (lane 2), EDTA-blood/stored for 2 days at 4°C (lane 3), EDTA-blood/frozen (lane 4), Citrate-blood/fresh (lane 5), Citrate-blood/stored for 2 days at 4°C (lane 6), Citrate blood/frozen (lane 7), Heparin-blood/fresh (lane 8) and Heparin-blood/frozen (lane 9). | Genomic DNA was prepared from blood to amplify a 1387bp fragment of the DMD-Gene (Duchenne Muscular Dystrophy). Increasing amounts of DNA template were used; 5ng (lane 2), 10ng (lane3), 25ng (lane 4), 50ng (lane 5), 75 ng (lane 6), 100ng (lane 7), 150ng (lane 8) and 200ng (lane9). Lane 10 is empty. Lane 11-18 show the results of amplification under the same conditions as lane 2-9 when a competitor product to prepare genomic DNA was used. Lane 1 is the length standard lambda HindIII/EcoRI. | |
