The Invitrogen 1kb ladder was run on a 1% TAE agarose gel. Single bands of this length standard were extracted using the JETQUICK Gel Extraction Spin Kit. The purified fragments were run again on a 1% TAE agarose gel.   EcoRI digested pBluescript II KS (2961bp) was extracted from a 1% TAE agarose gel using the JETQUICK Gel Extraction Spin Kit. Ligations were set up for 2 hours at 25°C with 0.5 to 3.0 units of T4 DNA ligase in 0.5U increments (lane 3-8). Lane 2 is the unligated plasmid DNA.
