Recombinant plasmid DNA (pT7T3 19U, 1.7kb insert) was prepared using the JETSPIN Plasmid Kit. The purified DNA was digested with various restriction enzymes, such as BamHI (lane 2), BglI (lane 3), DdeI (lane 4), DraI (lane 5), EcoRI (lane 6), HaeIII (lane 7), HindIII (lane 8), HpaII (lane 9), PvuI (lane 10), PvuII (lane 11), PvuI/PvuII (lane 12), RsaI (lane 13) and BglI/NdeI (lane 14). Lane 1 shows the undigested plasmid DNA.   Plasmid DNA (pT7T3 19U) was prepared using the JETSPIN Plasmid Kit. The plasmid DNA was digested with EcoRI and ligated with a 1.7kb DNA fragment (lane 2 shows the DNAs before ligation). Ligations were set up for 1h at 25°C with 0.03 U (lane 3), 0.06 U (lane 4), 0.08 U (lane 5), 0.1 U (lane 6) and 0.12 U (lane 7) of T4 DNA ligase per µg plasmid DNA. Length standard in lane 1 is the BRL 1kb ladder.
 
 
Midi and Maxi preparations:

The JETSPIN Plasmid Kits are offered with Midi and Maxi JETSPIN Columns for large-scale plasmid preparations. No vacuum manifold, special racks or any other additional auxiliaries are required. The spin columns can be used with any standard bench or floor model centrifuge with an appropriate rotor that can hold 50 ml centrifuge tubes (i.e. Falcon, Greiner, Sarstedt). A swinging-bucket type rotor is an absolute requirement. Centrifugations are carried out at low g-forces (5000 x g). Regardless of which JETSPIN Column format is used, the time of plasmid preparation is no more than 25 min.
